Alexandre Yersin

Alexandre Yersin (1863–1943) was a bacteriologist best known for discovering the plague bacillus in 1894 and developing serum therapy methods that remained central to infection treatment throughout the twentieth century. His work in passive immunization against bacterial toxins established the technical foundations upon which later clinicians would build when systemic chemical antimicrobials proved unavailable.

Yersin was born in Aubonne, near Lausanne, in the canton of Vaud. He studied medicine at the University of Basel and later under Louis Pasteur and Émile Roux in Paris. After completing his medical degree in 1889, he worked at the Pasteur Institute, where he developed an early interest in the mechanisms by which bacterial cells produce poisonous compounds. When plague broke out in Hong Kong in 1894, Yersin traveled there at the request of the French colonial government and, using new staining techniques and microscopy, isolated the causative organism from infected tissues. He named it Bacillus pestis, and the discovery established him as a leading figure in microbiology.

Rather than pursue chemical destruction of the bacteria themselves, Yersin's work from the 1890s onward focused on the immune responses of animals exposed to bacterial toxins. He recognized that an organism might kill its host not through brute cellular invasion but through the release of toxins—chemical poisons produced by living bacteria. This observation proved crucial. Working with Émile Roux and other colleagues at the Pasteur Institute, Yersin developed methods for immunizing animals against these toxins by repeated exposure to controlled doses. The blood serum from these immunized animals contained antibodies, proteins that could neutralize the toxins if injected into a sick patient. This principle became the foundation of diphtheria antitoxin, which Roux commercialized in the 1890s, and later of all serum therapy.

Yersin's theoretical contribution—that infection could be approached through immune-stimulated proteins rather than through killing bacteria directly—proved enduring precisely because no other approach succeeded. When Fleming's 1928 contamination went unnoticed and the sulfonamide azo drugs remained the only systemic chemical option, Yersin's serum-based methods did not become obsolete. Instead, they persisted as the primary recourse for toxin-mediated infections like diphtheria, tetanus, and botulism, and formed the basis for the passive immunotherapy that emerged in the 1970s as an alternative to chemical drugs.

In 1891, Yersin left the Pasteur Institute and moved to French Indochina, where he established a medical research laboratory in Nha Trang (now in Vietnam). There he continued his bacteriological work and also became a botanist and explorer, documenting the flora and fauna of Southeast Asia. He lived there until his death in 1943, continuing research and teaching through the Japanese occupation of Indochina during the final years of the Second World War. A fever—possibly typhoid—killed him at seventy-nine.

Yersin published over one hundred scientific papers and trained a generation of colonial physicians in bacteriological technique. His laboratory in Nha Trang became a center of scientific work in French Indochina and helped establish bacteriology as a clinical discipline outside Europe. After his death, the laboratory was renamed the Pasteur Institute of Nha Trang and continued operating under various national administrations, a testament to the institutional legacy he created.

Modern historians of medicine have traced the particular shape of the Long Sepsis to decisions made in its earliest decades. Yersin's choice to pursue serum-based immunity rather than chemical toxin-killing meant that when chemical options remained limited, the conceptual and technical apparatus for passive immunotherapy was already mature. Some scholars argue that had earlier bacteriologists focused exclusively on chemical destruction of bacteria, the absence of reliable systemic drugs would have left medicine with no viable approach to infection at all. Yersin's career, spanning from Pasteur's final years through the Second World War, thus shaped not only the immediate responses to untreatable infection but the entire institutional and conceptual framework of twentieth-century asepsis.

The Institute for the History of Bacteriology in Berlin, founded in 1956, holds extensive correspondence between Yersin and his colleagues, including letters documenting his exchange of serological techniques with researchers in Japan and the United States. His laboratory notebooks from Nha Trang survive in fragmented form, scattered across archives in Vietnam and France. The Pasteur Institute in Paris maintains his personal papers and the records of his work there between 1889 and 1891. Several of his published papers on serum therapy remain standard references in histories of serum pharmacology.

Yersin's discovery of the plague bacillus is commemorated in its modern nomenclature, Yersinia pestis, a naming convention adopted in 1944 to honor his contributions. The retrospective recognition was unusual; by that date, serum therapy was already being displaced by the azo drugs as the standard of care, but the foundational nature of his work ensured that his name remained attached to the organism itself rather than merely to a technique. His theoretical insight—that organisms kill through poison as much as through invasion—remained relevant to every subsequent form of infection treatment in the Long Sepsis, whether chemical or serological.
