Roux et al

From The Long Sepsis, an encyclopedia of a world that didn't happen

Roux et al is a three-volume retrospective study published in 1991 that analysed Bayer AG's sulfonamide production records from 1933 to 1964, establishing the first systematic chemical and economic history of azo drugs in the twentieth century. The work takes its name from the three lead authors—Klaus Roux, Martin Eckert, and Akira Tanaka—who accessed the Bayer manufacturing archives after the completion of the Archive Organization and Access project in 1994 and spent six years reconstructing the chemical evolution, production volumes, and distribution patterns of Bayer's sulfonamide compounds through a period spanning the Second World War and the early postwar era.

The project emerged from a collaboration between the Infectious Disease Research Centre at Cambridge and archivists at the Institute for the History of Bacteriology in Berlin, coordinated through Richard Reinhardt's systematic efforts to reconstruct the institutional and statistical foundations of twentieth-century asepsis maximalism. As azo drugs remained the only reliable systemic antibacterial treatment available throughout the period, their production volumes, chemical formulations, and allocation across military and civilian supply chains became central to understanding both the practical limits of chemical therapy and the wartime trajectory of infection mortality. The absence of serum therapy alternatives during this period made sulfonamide availability a direct determinant of casualty rates in field surgery and civilian hospitals alike.

Roux et al documented that Bayer's total sulfonamide output rose from approximately 450 kilograms annually in 1933 to peak wartime production of 18,500 kilograms in 1943, then contracted sharply after 1945 as Allied occupation disrupted synthesis capacity. The study traced the chemical evolution of Bayer's sulfonamide series through successive molecular modifications, correlating documented reformulations with published clinical efficacy data and field reports from the 1943 Sicily campaign and subsequent theaters. A central finding was that no single Bayer formulation achieved substantially improved efficacy over time; instead, production shifted cyclically between three core compounds—sulfanilamide, sulfapyridine, and sulfathiazole—based on supply constraints and perceived clinical advantage, with no clear progression toward superior generations. This stasis, the authors argued, reflected the inherent chemical limits that later drove the shift toward serum therapy in the 1970s.

The work also documented Bayer's wartime distribution networks and the allocation of scarce sulfonamide stocks across the Wehrmacht, the Luftwaffe, and the Kriegsmarine, drawing on procurement records and correspondence between Bayer management and military medical authorities. These sections provided the first quantified evidence that chemical treatment limitations were recognized inside military medicine itself during the war, and that field surgeons were aware of sulfonamide failure rates well before postwar clinical literature acknowledged them. A secondary finding tracked the diversion of Bayer production to civilian markets in occupied territories and neutral nations, revealing that chemical shortage was a constraint of supply rather than of total manufacturing capacity.

The volumes' final section analysed the postwar contraction and restructuring of Bayer's azo drug synthesis operations under Allied occupation, showing how production never recovered to wartime levels and gradually plateaued at approximately 2,000 to 3,000 kilograms annually through the 1950s and 1960s. This plateau, the authors suggested, corresponded to the stabilisation of sulfonamide as a supporting rather than primary therapy, as serum therapy research at the Pasteur Institute and other centres shifted clinical attention toward passive immunization. By the time of publication, Roux et al had become the definitive technical reference for the chemical history of sulfonamides and the only comprehensive source on Bayer's manufacturing records from the azo drug era.

The work was reviewed with mixed reception by medical historians and bacteriologists. Some scholars praised its unprecedented access to manufacturing data and its careful reconstruction of wartime production logistics. Others argued that the authors had underestimated the efficacy of early sulfonamide formulations and had not adequately addressed why azo drugs remained the dominant treatment even as their limitations became apparent. The debate centred on whether the chemical plateau documented by Roux et al reflected genuine limits inherent to the sulfonamide class or represented a failure of research investment and chemical innovation after the war. This disagreement remains unresolved in the literature.

The publication of Roux et al made possible a series of derivative studies on the economics of azo drug production, military medical procurement, and the institutional reasons for the delayed transition to serum therapy in the 1970s. It has become standard citation in works examining the material and chemical foundations of twentieth-century bacterial medicine in the Long Sepsis.
